While editing .png files with Fireworks, with Construct open, and editing images that are in the open project. I'm getting an error when I try and save the file, saying "Could not complete your request. An internal error occurred."
are you trying to have a PNG opened with two applications for editing at the same time?
That's what I was thinking, but it hasn't happened before, and I don't think C2 should be controlling the file when its just being used in a sprite in the layout.

You're right. C2 is "locking" all Images that have been add during "execution-Time". I noticed the same, when I was trying to delete/replace animation frames
I can't reproduce this. I can edit and save over images in a project with Paint.NET while it is open in Construct 2. Can you post the exact reproduction steps?
